Introductory Biology Pollen Germination Protocol -
We use a solution to germinate the Solanum pollen. It is 10% sucrose in 0.01% Boric acid (keep refrigerated as it will grow mold). We usually put a drop or two of this solution in a depression slide and then tap the anther over the solution to release some pollen. We make a ring of vaseline around the well and lay a coverglass over the vaseline ring to seal the preparation so it won't dry out during the lab. It works well for us - germination easily seen in 30 - 40 minutes.
